About This Product

What is retatrutide?

Retatrutide (LY3437943) is a lab-made peptide from Eli Lilly that acts on three hormone receptors at once: GLP-1, GIP and glucagon. Semaglutide acts on one of them and tirzepatide on two. The third, glucagon, is what sets retatrutide apart. It raises the energy the body burns, while the GLP-1 and GIP signals lower how much is eaten.

That is why it is called a triple agonist, or “triple G”. As of October 2026 it is an investigational drug, not an approved medicine anywhere.

How does retatrutide work?

  • GLP-1 receptor: releases insulin only when blood sugar is high, slows how fast the stomach empties and signals fullness to the brain.
  • GIP receptor: adds to the insulin response. In lab tests retatrutide is stronger at this receptor than at the other two.
  • Glucagon receptor: raises energy use and fat burning, mostly in the liver.

A fatty-acid chain on the peptide binds to albumin in the blood, which is what gives it a half-life of about six days in human studies.

What do researchers study it for?

  • Obesity and energy-balance models, with semaglutide and tirzepatide as the comparisons
  • Fatty liver research, where the glucagon signal shows most clearly
  • Receptor pharmacology: how three signals add up inside one molecule
  • Combination work with the amylin analogue cagrilintide
